Pharmacology

This is a combination of acetaminophen and caffeine. Paracetamol has analgesic and antipyretic properties, and its anti-inflammatory activity is weak. Caffeine is an alkaloid, a derivative of xanthine, similar to theophylline. Through the intermolecular association with paracetamol, caffeine increases the solubility and transmembrane permeability of paracetamol. In addition, caffeine increases pain threshold and pain tolerance. Caffeine also has the inherent power to increase the tone of blood vessels in the brain, which provides another benefit for the treatment of migraines and headaches.

Dosage & Administration

Adult dose: 1-2 tablets every 4-6 hours. Maximum dose: 8 tablets daily.

Child dose: Not recommended for children below 12 years.

Interaction

You can use anticonvulsants (such as phenytoin, barbiturates, carbamazepine) to reduce serum levels. Long-term use can enhance the blood thinning effect of warfarin and other coumarins. It accelerates absorption together with metoclopramide and domperidone. You can use probenecid to increase serum levels. May increase the serum level of chloramphenicol. You can reduce the absorption within 1 hour after taking cholestyramine. The use of phenothiazines may cause severe hypothermia.

Contraindications

Acetaminophen is contraindicated in patients with severe renal failure and liver disease (viral hepatitis). Known to be allergic to paracetamol or caffeine.

Side Effects

The side effects of paracetamol are usually mild, but haematological reactions including thrombocytopenia, leukopenia, pancytopenia, neutropenia, and agranulocytosis have been reported. Occasionally, pancreatitis, skin rash, and other allergic reactions occur.

Pregnancy & Lactation

Pregnant women should consult a doctor before taking acetaminophen and caffeine. You can take acetaminophen and caffeine while breastfeeding.

Precautions & Warnings

Paracetamol and caffeine should be used with caution in the following situations: Patients with liver or kidney failure, patients taking other hepatotoxic drugs. Avoid using the drug for a long time without consulting a doctor.
